We actually only get to see the soul stone used once in the entire movie, and you may not have noticed when this was. When the heroes that are on Titan are fighting against Thanos, at one point Doctor Strange makes multiple versions of himself to take on the Mad Titan. During this moment, look closely at the infinity gauntlet when Thanos fights back. You will see that the soul stone lights up. That is because Thanos uses the stone to attack the real Strange, which is why after the attack, Strange’s soul leaves his body for a moment.
